乙酰丙酮分光光度法测定白菜中甲醛含量的方法优化及其在快检结果验证中的应用
目的 建立乙酰丙酮分光光度法测定白菜中甲醛含量的方法, 并用此方法验证甲醛速测盒的检测结果。方法 以甲醛含量为指标, 研究样品浸泡时间、加蒸馏水量、加磷酸量、蒸馏时间等前处理条件和显色时间、水浴时间、加乙酰丙酮量等测定条件对白菜中甲醛测定结果的影响。以大白菜为试样, 对比乙酰丙酮分光光度法和甲醛速测盒的检测结果。结果 白菜中甲醛含量测定的最佳的条件为样品浸泡时间35 min, 加水量160 mL, 磷酸添加量5 mL, 蒸馏时间80 min, 乙酰丙酮添加量1 mL, 水浴时间5 min, 显色时间15 min。在此条件下, 白菜中甲醛回收率为95.2%~99.7%。甲醛速测盒的结果与之比对, 结果一致。结论 乙酰丙酮分光光度法结果准确, 适合用于大白菜中甲醛的测定, 实验中所用甲醛速测盒与之结果一致, 可以快速有效测定白菜中甲醛含量。
Objective To establish a method for the determination of formaldehyde in cabbage by acetylacetone spectrophotometry, and to verify the detection results of the formaldehyde rapid test kit by this method. Methods Taking the formaldehyde content as the index, the effects of pretreatment conditions such as sample soaking time, amount of distilled water, amount of added phosphoric acid, distillation time, and the determination condition such as color development time, water bath time and amount of acetylacetone on the determination of formaldehyde in cabbage were studied. The Chinese cabbage was used as a sample to compare the detection results of acetylacetone spectrophotometry and formaldehyde rapid test kit. Results The optimum conditions for the determination of formaldehyde in cabbage were as follow: sample soaking time 35 min, water addition 160 mL,phosphoric acid addition 5 mL, distillation time 80 min, acetylacetone addition 1 mL, water bath time 5 min, color development time 15 min. Under these conditions, the formaldehyde recovery rate in cabbage was 95.2%-99.7%. The results of the formaldehyde rapid test box were compared and the results were consistent. Conclusion The method of acetylacetone spectrophotometry is accurate, which is suitable for the determination of formaldehyde in Chinese cabbage. The results of formaldehyde rapid test kit used in the experiment are consistent with the results of cetylacetone spectrophotometry, which can determinate formaldehyde in Chinese cabbage quickly and effectively.
