Objective To establish a method for simultaneous determination of 17 amino acids in quail eggs by microwave-hydrolyzed ninhydrin column post-derivative ion exchange chromatography. Methods The sulfonium-type cationic resin ion exchange column was used for separation, and the ninhydrin solution was used as a post-column derivatizer. The UV-visible spectrophotometer was used for detection. The detection wavelengths were 570 nm and 440 nm, and the microwave hydrolysis time and temperature were optimized. Results The 17 amino acids showed a good linear relationship in the range of 0.005?1.000 μmol/mL, and the correlation coefficient was greater than 0.99. The limits of detection were 0.376?0.906 mg/L. The results of microwave hydrolysis at 150 ℃ for 20 min were basically the same as those of traditional heating hydrolysis (110 ℃, 22 h). Conclusion This method has short analysis time and high sensitivity, and can be used for detecting the amino acid content in quail eggs.
