Objective To establish a sensitive colloidal gold immunochromatography method for the rapid detection of rhodamine B (RB) in pepper products. Methods Colloidal gold was prepared by sodium citrate reduction method, and it was labeled with RB monoclonal antibody by physical adsorption method. Under optimized conditions, a sensitive colloidal gold immunochromatography (GICA) was developed based on competitive binding immunoassay. Results RB in pepper products was detected by the established method, and the qualitative detection limit was 5 μg/kg, which met the requirements of national security limits. The results of the actual samples by this method were consistent with those of the liquid chromatography-mass spectrometry (LC-MS) method. Conclusion The established GICA is sensitive, rapid and efficient for the illegal use of RB residues in chili products.
