灵芝多糖脱色工艺研究
目的 优化灵芝多糖脱色工艺，选取最优脱色方式。方法 选取活性炭，壳聚糖，H2O2 3种方法对灵芝多糖脱色处理，以脱色率和多糖保留率为指标，在单因素实验基础上，进行正交工艺优化。以2,2-二(4-叔辛基苯基)-1-苦肼基自由基(DPPH·)清除活性分析抗氧化活性。结果 活性炭对灵芝多糖的脱色率为67.72%，多糖保留率为72.12%；壳聚糖对灵芝多糖的脱色率为33.57%，多糖保留率63.00%；H2O2对灵芝多糖的脱色率为84.11%，多糖保留率为73.12%，综合考虑H2O2去色素效果最好。3种方式对DPPH都具有一定的清除能力，由EC50值可知壳聚糖脱色法所得多糖的DPPH自由基清除能力最强。结论活性炭，壳聚糖，H2O2 3种方法均可脱色素，H2O2效果较好，值得进一步的开发和利用。
Objective To optimize the decolorization process of Ganoderma lucidum polysaccharides and select the best decolorization method. Methods Activated carbon, chitosan and H2O2 were selected for decolorization, and the decolorization rate and polysaccharide retention were used as indicators. On the basis of single-factor experiment, to perform orthogonal process for optimization. Their antioxidant capacities were investigated according with scavenging of 2,2-diphenyl-1-picrylhydrazyl(DPPH·) radicals. Results The decolorization rate of activated carbon for ganoderma polysaccharides was 67.72%, the polysaccharide retention rate was 72.12%; the decolorization rate of chitosan for ganoderma polysaccharides was 33.57%, the polysaccharide retention rate was 63.00%; the decolorization rate of H2O2 for ganoderma polysaccharides was 84.11%, and the polysaccharide retention rate was 73.12%. All of the 3 kinds of methods had certain scavenging capacity for DPPH. According to the EC50 value, the polysaccharides obtained by chitosan depigmentation method had the strongest scavenging capacity for DPPH free radical. Conclusion Activated carbon, chitosan and H2O2 can be used for depigmentation, and the effect of H2O2 is much better, which is worthy to further development and utilization.
