正交设计法优化富硒发芽糙米中硒蛋白提取工艺
目的 优化硒蛋白的分离提取技术。方法 以富硒发芽糙米为原料, 采用碱提酸沉法提取硒蛋白, 研究NaOH溶液浓度、料液比、浸提时间、pH值、(NH4)2SO4饱和度5个因素对硒蛋白产量的影响, 并通过正交试验确定硒蛋白分离提取的最优工艺。结果 硒蛋白分离提取的最佳条件为: NaOH溶液浓度0.12 mol/L、料液比1:10(g:mL)、浸提时间4 h、pH 4.5、(NH4)2SO4饱和度50%。在此条件下, 10 g富硒发芽糙米中硒蛋白产量为327.56 mg, 即硒蛋白得率约为3.28%, 硒含量为0.73 μg, 换算成硒得率为48%。结论 本方法简便快捷且硒蛋白及硒得率较高, 可作为富硒发芽糙米中硒蛋白的分离提取方法。
Objective To optimize the separation and extraction technology of selenoprotein. Methods With selenium enriched germinated brown rice as raw material, the selenoprotein was extracted by alkaline acid precipitation method. Effects of NaOH solution concentration, ratio of material to liquid, extraction time, pH value and (NH4)2SO4 saturation on the yield of selenoprotein were studied, and the optimal process of selenoprotein extraction was determined by orthogonal test. Results The optimal conditions for the separation and extraction of selenoprotein were as follows: NaOH solution concentration was 0.12 mol/L, material liquid ratio was 1:10(g:mL), extraction time was 4 h, pH was 4.5 and (NH4)2SO4 saturation 50%. Under this condition, the yield of selenoprotein in 10 g selenium germinated brown rice was 327.56 mg, the selenoprotein yield is about 3.28% and the selenium content was 0.73 μg, converting into selenium yield was 48%. Couclusion The method is simple, rapid and has high selenoprotein and selenium yield, which can be used as a separation and extraction method of selenoprotein in se-enriched germinated brown rice.
