高效液相色谱法测定维生素片中维生素B12的不确定度评定
目的 评定高效液相色谱法测定维生素片中维生素B12的不确定度。方法 维生素片中维生素B12经纯化水超声提取, 经色谱柱分离, 流动相等度洗脱, 使用紫外检测器550 nm检测, 外标法定量来测定维生素片中维生素B12的含量。依据JJF 1059.1-2012《测量不确定度评定与表示》, 考察称量、标准溶液配制、样品和仪器重复测量等主要因素引入的不确定度, 并对不确定度的各分量进行计算和合成。结果 当维生素片中维生素B12含量为0.408 mg/100 g 时, 在95%的置信区间下, 其扩展不确定度为±0.0053 mg/100 g(k=2)。评定结果表明, 实验过程中的不确定度主要来源于标准溶液配制、仪器重复测量。结论 不确定度评定适用于高效液相色谱法快速测定维生素片中维生素B12含量的不确定度分析, 对检测结果准确度的提高具有指导 意义。
Objective To evaluate the uncertainty of the determination of vitamin B12 in vitamin tablets by high performance liquid chromatography (HPLC). Methods Vitamin B12 was extracted by purified water, separated by chromatographic column, and eluded by flow equality, 550 nm UV detector and the external standard method was used to determine the vitamin B12 content in vitamin tablets. According to JJF 1059.1-2012 Measurement uncertainty evaluation and presentation, the uncertainty of main factors such as weighing, standard solution preparation, sample and instrument repeatability measurement were investigated, and the components of uncertainty were calculated and synthesized. Results When the vitamin B12 content was 0.408 mg/100 g, the extended uncertainty was ±0.0053 mg/100 g(k=2) under the 95% confidence interval. The evaluation results showed that the uncertainty of the experiment was mainly derived from the standard solution and the repeated measurement of the instrument. Conclusion The uncertainty assessment is suitable for the determination of vitamin B12 in vitamin tablets by HPLC, and it is instructive to improve the accuracy of test results.
