高效液相色谱法检测不同采收期邵阳龙牙百合的秋水仙碱含量
目的  建立高效液相色谱法比较不同采收期邵阳龙牙百合中秋水仙碱含量。方法  样品加水浸泡过夜后超声提取, 再用氯仿萃取, 氯仿层水浴蒸干, 以流动相溶解稀释。色谱柱: Waters C18(250 mm×4.6 mm,   5 μm); 流动相: 甲醇-水(40:60, V:V), 等度洗脱; 流速: 1.0 mL/min; 检测波长: 254 nm; 柱温: 30 ℃; 进样量: 20 μL。结果  秋水仙碱在1.025~20.50 μg/mL范围内线性关系良好(r2=0.9997, n=5), 回收率为99.2%(n=9)。样品实测显示, 不同采收期龙牙百合秋水仙碱含量不同, 茎叶枯黄脱落时期含量最高, 平均含量达到3.64 mg/g。结论  该法简单快速, 定量准确, 可以为评价龙牙百合质量和选择最佳采收时间提供参考。

Objective  To establish a method for determination of colchicine in Lilium brownii var. viridulum with different harvest time by high performance liquid chromatography. Methods  The samples were ultrasonic extracted after soaking in the water for the night, then extracted with chloroform. The chloroform layer was steamed with water bath, and the residue was dissolved and diluted by flow phase. Column was Waters C18 (250 mm×4.6 mm, 5 μm), mobile phase was methanol-water (40:60, V:V), flow rate was 1.0 mL/min, detection wavelength was 254 nm, column temperature was 30 ℃, and sample amount was 20 μL. Results  The colchicine showed a good linear relationship within the range of 1.025~20.50 μg/mL (r2=0.9997, n=5), and the average recovery was 99.2% (n=9). The results of sample measurement showed that the content of colchicine in Lilium brownii var. viridulum was different in different harvest time, which stems and leaves turn yellow and fall off period was the highest, and the average content of colchicine in Lilium brownii var. viridulum reached 3.64 mg/g. Conclusion  This method is accurate, simple and reliable, which provides a reference for the evaluation of quality and selection of the best harvest time of Lilium brownii var. viridulum.
