高效液相色谱-串联质谱法同时测定食品中9种人工色素
目的 建立一种同时测定食品中9种人工色素(赤藓红、诱惑红、柠檬黄、亮蓝、靛蓝、胭脂红、苋菜红、日落黄、新红)的高效液相色谱-串联质谱(high performance liquid chromatography-tandem mass spectrometry, HPLC-MS/MS)检测方法。方法 3种食品样品分别经3种不同的固相萃取柱净化方法净化, 采用甲醇(A)和10 mmol/L乙酸铵(B)为流动相, 梯度洗脱分离, 电喷雾离子化, 正离子扫描, 多反应监测对9种人工色素进行定性和定量。结果 本方法在15 min内完成9种目标化合物的分离分析; 采用PLS固相萃取小柱进行前处理, 各色素在100 μg/kg添加水平的回收率为98.1%~102.3%, 结论 该前处理方法回收率高, 仪器方法快速、准确、灵敏, 适合测定食品中9种人工色素。
Objective To establish a method for simultaneous determination of 9 kinds of synthetic colorants (erythrosine, allura red, lemon yellow, brilliant blue, indigo, carmine, amaranth, sunset yellow and new red) in food samples by high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). Methods Totally 3 samples were extracted by 3 different solid phase extraction methods, and then 9 kinds of synthetic colorants in the extracts were separated on a reversed phase using a gradient elution program of methanol (A) and 10 mmol/L ammonium acetate (B) . Electrospray positive ionization (ESI+) was used with multiple reactions monitoring (MRM), and the major components of the 9 kinds of synthetic colorants were performed based upon the intensities of fragments. Results The major components of the 9 kinds of synthetic colorants were analyzed within 15 min by the established method and the recoveries were ranged from 98.1% to 102.3% for the 9 kinds of synthetic colorants with 100 μg/kg by PLS solid phase extraction method. Conclusion The proposed method has high recoveries, and is rapid, accurate and sensitive, which is suitable for detecting 9 kinds of synthetic colorants in food samples.
