呋喃唑酮代谢物时间分辨荧光免疫快速检测试剂卡的研制及应用
目的 应用时间分辨免疫荧光检测技术, 研制出一种快速检测呋喃唑酮代谢物的试剂卡。方法 采用EDC介导法偶联抗体, 由于铕荧光微球的荧光持续稳定性好, 强度大, 不易受干扰等优点, 将其作为荧光标记物。通过优化铕荧光微球与单抗之间的用量, 改善试剂卡的显色梯度, 提高试剂卡的灵敏度。结果 结果显示本试剂卡对呋喃唑酮代谢物的检出限为0.5 μg/kg, 与呋喃唑酮代谢物类似物的交叉反应率低, 可在室温条件下保存一年; 此方法下的检测结果与液相色谱-串联质谱法的检测结果符合程度为100%。结论 呋喃唑酮代谢物时间分辨荧光免疫快速检测试剂卡具有检测快速、准确、灵敏度高的特点, 操作简单, 能满足大规模现场快速检测的需求, 为食品安全每个环节提供保障和便利。
Objective To develop a reagent card for the rapid detection of furazolidone metabolites by using time-resolved immunofluorescence detection. Methods Using EDC mediated method coupled antibody, the europium fluorescent microspheres were selected as fluorescent marker due to its advantages, such as high fluorescence persistent stability, high strength and strong anti-jamming-power. The dosage between europium fluorescent microspheres and monoclonal antibody was optimized to improve the strips’ chromogenic gradient and sensitivity. Results The results showed that the detection limit of furazolidone metabolite by this reagent card was 0.5 μg/kg. The cross-reaction rate of furazolidone metabolites was low and could be preserved at room temperature for one year. The test results under this method were in agreement with the results of liquid chromatography-tandem mass spectrometry. Conclusion The rapid detection reagent strip for furazolidone metabolite residue has the characteristics of simple operation, rapid speed, accuracy and high sensitivity. It can meet the needs of large-scale on-site rapid testing, and provide guarantee and convenience for every link of food safety.
