夏枯草水提物对吡唑-脂多糖诱导肝损伤的 保护作用
目的 研究夏枯草水提物对吡唑-脂多糖诱导的小鼠肝脏损伤的保护作用。方法 将42只小鼠随机分为6组: 正常对照组、模型组、夏枯草水提物低、中、高剂量组以及阳性对照组。灌胃给予小鼠夏枯草水提物, 同时采用腹腔注射吡唑-脂多糖造成小鼠肝损伤模型, 检测血清谷丙转氨酶(ALT)、谷草转氨酶(AST)和肝组织过氧化氢(H2O2)、丙二醛(MDA)、过氧化氢酶(CAT)、总超氧化物歧化酶(T-SOD)和谷胱甘肽(GSH)的活性/含量以及细胞色素酶CYP2E1和CYP2A5的蛋白表达水平。结果 吡唑-脂多糖能够引起提高CYP2E1和CYP2A5的蛋白表达以及肝脏氧化应激并造成肝损伤; 夏枯草水提物能够显著降低CYP2E1和CYP2A5的蛋白表达, 降低氧化产物H2O2和MDA的含量, 以及提高抗氧化系统中CAT、T-SOD活性和GSH含量。结论 研究表明夏枯草可能通过抑制CYP2E1和CYP2A5的蛋白表达降低吡唑-脂多糖诱导肝组织内的氧化应激, 从而对吡唑-脂多糖引起的肝损伤起保护作用。
Objective To explore the protective effect of Prunella vulgaris L. (PV) against pyrazole-lipopolysaccharide (LPS) induced liver injury in mice. Methods A total of 42 mice were randomly divided into 6 groups: normal control group, model group, positive control group and low, middle and high dose of PV group. Mice were treated with PV by oral gavage, and liver injury of model mice were treated with pyrazole-LPS by the injection. The liver function was evaluated by detecting the activity of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), and hepatic hydrogen peroxide (H2O2), catalase (CAT), total super-oxidedismutase (T-SOD), glutathione (GSH), malondialdehyde (MDA), and the protein expression of CYP2E1 and CYP2A5. Results Pyrazole-LPS could induce liver injury by increase the CYP2E1 and CYP2A5 protein levels and oxidative stress. PV extraction could reduce the CYP2E1 and CYP2A5 protein levels, which led to the reduction of H2O2 and MDA as well as the enhancement of anti-oxidative defense (CAT, T-SOD and GSH). Conclusion These findings showed the hepato-protective properties of PV against pyrazole-LPS induced liver injury by attenuating the protein levels of CYP2E1 and CYP2A5, and enhancing the protective role of anti-oxidative defense system.
