OBJECTIVE:Ratmodelsofhypobarichypoxia-inducedpulmonaryhypertensionarecommonlyusedinstudiesofchronicmountainsickness,whiletherearefewresearchesspeciallyfocusingontheseratsmodel.Thisstudyaimstoexploringpossiblepathogenesisofhypobarichypoxia-inducedpulmonaryhypertensionbyexperimentingonhypobarichypoxia-inducedPHratmodelsatdifferentsimulate-altitudes.MATERIALSANDMETHODS:32healthymaleSDratswererandomlydividedintosixgroupsofdifferentdegreeandtimeperiodofhypobarichypoxia.Themeanpulmonaryarterialpressure(mPAP),rightventricularpressure(RVSP),therightventricle(RV),leftventricular(LV),ventricularseptal(S),therightventricularhypertrophyindex(RVHI)[calculatedundertheformulaofRV/(LV+S)],hematoxylin-eosinstaining,elasticfibersstaining,theratioofthethicknessofvascularwalltoitsouterdiameter(MT%),theratioofthecross-sectionalareaofthemiddlevascularwalltothetotalvascularcross-sectionalarea(MA%);theα-SMA,andtheKi6expressionsweredetectedtoevaluatedthepulmonaryhypertension.RESULTS:ThereweresignificantdifferencesofthemPAP,RVSPandRVHIvaluebetweenthehypobarichypoxiagroupsandthecontrolgroup(p<0.05).ThemPAP,RVSP,RVHI,MT%,MA%,α-SMA,andKi6ofratsinmodelgroupsatanaltitudeof3KMwerehigherthanthoseofthecontrolgroup,whichraisedgraduallywiththenumberofweeksincreasing.ThemPAP,RVSP,RV/(LV+S)value,MT%,MA%,α-SMA,andKi67ofthe5KM-4Wgroupweresignificantlyhigherthanthoseofthecontrolgroup(p<0.05).CONCLUSIONS:Ratmodelswithpulmonaryhypertensionatdifferentaltitudeshavebeensuccessfullyestablishedbyautomaticadjustinghypobarichypoxiachamber.Exposuretoalowoxygenenvironmentatasimulate-altitudeof3kmfor8weekshavecausedthepathologicalremodelingofpulmonaryvascularwallsandpulmonaryhypertension,andfurtherledtoaseriesofpathologicalchanges,includingrightventricularhypertrophy.Thismodeliseasytoberepli-catedwithgoodreproducibilityandprovidesevidenceforclinicaltrialofdrugs.
