单测试片法定性快速检测乳中金黄色葡萄球菌的研究
目的建立单测试片定性快速检测乳中金黄色葡萄球菌的方法。方法选择7.5%氯化钠肉汤增菌液, 使用拍打式均质器, 匀浆时间1~1.5 min进行样品处置。采集不同基质样品, 设计多种方案对研究单测试片方法、国标方法与测试片方法同时进行验证。结果该方法检测金黄色葡萄球菌特异性高, 特异性达到96.7%。最小检出量0.5 CFU/mL。阳性检出率高。结论该方法准确性可靠, 特异性、灵敏度都高, 检出限低, 适用于乳中的金黄色葡萄球菌快速定性检测。
ObjectiveTo establish a method of rapid qualitative detection of Staphylococcus aureus in milk by single test method. Methods7.5% sodium chloride broth enrichment solution was selected, a tapping homogenizer was used, and the sample was homogenized for 1?1.5 min. Different matrix samples were collected and various schemes were designed to verify the single test method, national standard method and test piece method at the same time. ResultsStaphylococcus aureus was highly specific and had a specificity of 96.7%. The minimum detectable amount was 0.5 CFU/mL. The positive detection rate was high. ConclusionThe method has high accuracy, specificity, sensitivity, and low detection limit, it is suitable for the rapid qualitative detection of Staphylococcus aureus in milk.
