Breastcanceristhemostcommoninvasivecancerinfemalesacrosstheworld,whosemortalitycouldbeseriouslyaffectedbyearly-detectionandprecisetherapy.AsaFDA-approvedcontrastagentforopticalimagingtogetherwiththeabilityforPTT/PDT,ICGhasattractedgreatresearchattentionandachievedrapiddevelopmentinthepastdecades.Inthisresearch,weusedamphiphilicpeptideC18GR7RGDSasananocarriertoincorporateICGviasimplesonicationanddialysis.TheformedPEP-ICGwascharacterizedbyDLSandTEM,suggestingasphericalstructurewithdiametersof134nminwater.ThesuccessfulencapsulationofICGwereconfirmedandanalyzedwith30％loading.TheabilityofphotothermalconversionandROSproductionwerecomparableandevenbettercomparedtoICGalone.Thecytotoxicitywith/withoutlaserandcellularuptakeofPEP-ICGwereevaluatedonbothαvβ3+MDA-MB-231cellsandαvβ3-MCF-7cellsviaCCK-8,confocalfluorescencemicroscopyandflowcytometry.TheresultsshowedthatPEP-ICGcouldspecificallytargetMDA-MB-231.Moreover,PEP-ICGwithgreatbiocompatibilitybecametoxiconceexposedtoNIRlaser.TheexcellentintracellularphotodynamiceffectofPEP-ICGwasalsoverifiedbyconfocal.Inconclusion,PEP-ICGnanomicellesasatheranosticnanoplatformhavedemonstratedexcellentutilityandgreatpotentialsfortheearly-detectionandprecisetherapyofbreastcancers.
